Filed a tax return how long does it usually take to get some kind of response? ???

Do you mean when do you get a refund? I think there might be a way to track your refund online. If you have to pay taxes, you won't get a response; you will just see the amount deducted from your bank account.

Just got a letter stating I owe 400 pounds and unfortunately I didn't mark on my self assessment that I wanted it taken out of my PAYE (adjusted I guess is the right word) is it possible for me to still request this?

I don't believe so, if its HMRC that's sent the letter to you you have to pay them upfront yourself by their deadline before they add late charges on top of it.   :-\\\\
